以香稻1号为材料,对64对PstⅠ-MseⅠ选扩引物进行筛选,确定19对引物能扩增出适量且清晰的条带。以19对引物对具有不同表型性状的4种T5代变异株系及其对照进行AFLP分析,结果表明,变异株系基因组较香稻1号发生了明显变化。回收克隆了变异株06-5-114-11-2的p38-114特异片段和06-5-108-11-3的p82-108特异片段,并在NCBI数据库中进行序列比对,结果显示,p38-114片段与序号为NM_001074855.1的mRNA序列相似性为99%,p82-108片段与序号为NM_001061652.1的mRNA序列相似性为100%。
Sixty-two PstⅠ-MseⅠsequencing primers were screened using Xiangao 1 as material, and 19 pairs of primers were able to amplify a moderate amount of clear bands. The AFLP analysis of four T5 generation mutant lines with different phenotypic traits and their controls with 19 pairs of primers showed that the genome of the mutant strain had a significant change compared with that of Xiangdao 1. The specific fragment of p38-114 and the specific fragment of p82-108 of 06-5-108-11-3 of the mutant 06-5-114-11-2 were cloned and sequenced in the NCBI database. The similarity of the mRNA sequence of p38-114 with the sequence number NM_001074855.1 was 99%. The sequence similarity of the p82-108 fragment with the sequence number NM_001061652.1 was 100%.
