采用RT_PCR技术 ,从豌豆 (PisumsativumL .)幼叶中克隆了 1个约 80 0bp的Lhcb2cDNA。以特异探针进行的Southern杂交结果表明 ,Lhcb2基因以单拷贝形式存在于豌豆基因组中。不同光照时间和温度对豌豆幼苗进行处理的RT_PCR和Northernblotting分析表明 ,Lhcb2基因转录水平上的表达受光照的控制 ,且明显地表现出对光照时间的依赖性。光照 0~ 1.5hLhcb2基因未见表达 ,而光照 2h以上该基因才大量表达 ;温度对Lhcb2基因的表达亦有显著的影响 ,相同的光照处理 ,4℃下Lhcb2基因的表达量比 2 5℃下的表达量低 1倍左右。
Using RT_PCR technique, we cloned 1 80 bp Lhcb2 cDNA from young leaves of pea (Pisum sativum L.). Southern hybridization with a specific probe showed that the Lhcb2 gene is present in a single copy in the pea genome. RT_PCR and Northern blotting analysis of pea seedlings treated with different illumination time and temperature showed that the expression of Lhcb2 gene at the transcriptional level was controlled by light and obviously showed the dependence of illumination time. The expression of Lhcb2 gene was not observed in light at 0 ~ 1.5h, but was up-regulated at 2h after light irradiation. Temperature also had a significant effect on the expression of Lhcb2 gene. Under the same light treatment, the expression of Lhcb2 gene at 4 ℃ was lower than that at 25 ℃ The expression level is about 1 times lower.
