目的 分析转染醛糖还原酶相似基因 1(ARL 1)后 ,实验组人肝癌细胞与对照组细胞在基因表达谱方面的差异。 方法 以实验组及对照组细胞的总RNA反转录合成的cDNA为探针 ,与cDNA芯片 (cDNAchips)进行差异杂交 ,并应用半定量RT PCR对差异表达的重要的相关基因进行初步筛选。 结果 cDNA芯片分析发现 6种基因的表达水平明显上调 ,9种基因的表达水平明显降低 ,涉及细胞的蛋白质代谢、信号传导、转移和耐药性等方面。 结论 cDNA芯片分析发现 :转染醛糖还原酶相似基因 1后肝癌细胞的基因表达谱发生改变 ,为系统研究肝癌的耐药性及其机制提供了有益的线索。
Objective To analyze the gene expression profiles of human liver cancer cells and control cells transfected with aldose reductase similar gene 1 (ARL 1). Methods The cDNAs were reverse transcribed from the total RNA of the experimental group and the control group, and then were differentially expressed with cDNAchips. Semi-quantitative RT-PCR was used to screen the differentially expressed genes. Results The results of cDNA microarray showed that the expression levels of six genes were significantly up-regulated and the expression levels of nine genes were significantly decreased, which involved in the aspects of protein metabolism, signal transduction, metastasis and drug resistance. Conclusion The results of cDNA microarray analysis showed that the gene expression profile of hepatocellular carcinoma cells was changed after aldose reductase gene 1 transfection, which provided useful clues for systematically studying the drug resistance of liver cancer and its mechanism.
