目的探讨乳腺癌细胞株与乳腺癌临床标本中Smad4及转化生长因子(TGF)-β1、TGF-β受体(R)1的蛋白表达水平与乳腺癌发生、发展、转移等生物学行为及预后的相关性。方法利用免疫组织化学检测30例乳腺癌临床标本Smad4和TGF-β1、TGF-βR1蛋白表达,免疫细胞化学检测MCF-7、T47D、SKBR-3三种乳腺癌细胞的TGF-β1、TGF-βR1和Smad4蛋白表达,Western印迹检测乳腺癌TGF-βR1和Smad4蛋白表达,分析三种乳腺癌细胞TGF-β1、TGF-βR1和Smad4蛋白表达情况。结果免疫组化法结果显示,Smad4和TGF-βR1的表达水平在低度恶性的MCF-7细胞中明显高于高度恶性的SKBR-3细胞,中度恶性的T47D细胞介于二者之间。病理类型非浸润性癌组,TGF-β1、TGF-βR1及Smad4的表达水平明显高于恶性程度高的浸润性癌患者组,早期浸润性癌组介于二者之间。Western印迹显示,恶性程度低的MCF-7细胞,其TGF-βR1和Smad4的蛋白含量明显高于恶性程度高度的SKBR-3细胞。恶性程度低的患者TGF-βR1和Smad4的蛋白含量明显高于恶性程度高的患者。结论 TGF-β/Smads信号传导通路活性与乳腺癌的恶性程度呈负相关,Smad4、TGF-β1、TGF-βR1可能在乳腺癌演变中起抑制作用。
Objective To investigate the protein expression of Smad4, transforming growth factor (TGF) -β1 and TGF-β receptor (R) 1 in breast cancer cell lines and breast cancer and the biological behavior and prognosis of breast cancer Relevance. Methods The expression of Smad4, TGF-β1 and TGF-βR1 in 30 specimens of breast cancer were detected by immunohistochemistry. The expressions of TGF-β1 and TGF-βR1 in MCF-7, T47D and SKBR-3 breast cancer cells were detected by immunocytochemistry And Smad4 protein expression. Western blotting was used to detect the expression of TGF-βR1 and Smad4 in breast cancer cells. The expressions of TGF-β1, TGF-βR1 and Smad4 in breast cancer cells were analyzed. Results The results of immunohistochemistry showed that the expression of Smad4 and TGF-βR1 in MCF-7 cells with low malignancy was significantly higher than that in SKBR-3 cells with high grade, while the moderately malignant T47D cells were in between. The expression of TGF-β1, TGF-βR1 and Smad4 in pathological type non-invasive cancer group was significantly higher than that in invasive group with high degree of malignancy, and early invasive cancer group was in between. Western blotting showed that the protein content of TGF-βR1 and Smad4 in MCF-7 cells with low degree of malignancy was significantly higher than that in SKBR-3 cells with high degree of malignancy. Patients with low grade of malignancy TGF-βR1 and Smad4 protein content was significantly higher than the high degree of malignancy in patients. Conclusion The activity of TGF-β / Smads signaling pathway is negatively correlated with the malignancy of breast cancer. Smad4, TGF-β1 and TGF-βR1 may play an inhibitory role in the development of breast cancer.
