目的研究表皮生长因子(enidermal growth factor, EGF)、神经降压素(neurotensin, NT)对四氯化碳损伤的原代培养肝细胞Ca~(2+)及膜流动性的影响。方法以Fura-2/AM和DPH荧光探针测定肝细胞内游离钙和膜流动性,观察EGF、NT对四氯化碳损伤后肝细胞这两个指标的影响。结果EGF、NT明显对抗肝细胞四氯化碳损伤后胞内[Ca~(2+)] i的增高和膜流动性的降低。结论 EGF、 NT有维持肝细胞膜流动性和胞浆钙稳态的作用。
Objective To investigate the effect of enotaxial growth factor (EGF) and neurotensin (NT) on Ca 2+ and membrane fluidity in primary cultured hepatocytes injured by carbon tetrachloride. Methods The intracellular free calcium and membrane fluidity were measured with Fura-2 / AM and DPH fluorescent probes. The effects of EGF and NT on the two indexes of liver cells after carbon tetrachloride injury were observed. Results EGF and NT obviously antagonized the increase of intracellular [Ca2 +] i and the decrease of membrane fluidity after carbon tetrachloride injury in hepatocytes. Conclusion EGF and NT can maintain the membrane fluidity and cytoplasmic calcium homeostasis in hepatocytes.
