为了探讨在发育过程中小鼠颌下腺神经生长因子(NGF)基因表达的变化,应用原位杂交组织化学技术和图像分析方法对新生至2月龄ICR雄性小鼠颌下腺NGFmRNA的相对含量及分布进行研究。结果显示:新生小鼠颌下腺未见NGFmRNA杂交信号,8~10d龄小鼠颌下腺NGF原位杂交信号呈阴性至弱阳性。12~14d龄小鼠颌下腺NGF原位杂交信号呈弱阳性,分布于纹状管上皮细胞胞浆中。1月龄小鼠颌下腺纹状管和颗粒曲管NGF原位杂交信号均呈阳性,位于细胞核周围胞浆中。2月龄小鼠颌下腺纹状管和颗粒曲管NGF原位杂交信号呈强阳性,主要分布在上皮细胞核下部胞浆。实验结果提示,雄性小鼠颌下腺NGF基因在生后1~2周龄间开始转录,NGFmRNA含量在2月以前随年龄的增长而增多
In order to investigate the changes of NGF gene expression in mouse submandibular gland during development, the relative content and distribution of NGF mRNA in submandibular gland of neonatal to 2-month old ICR male mice were studied by in situ hybridization and image analysis. The results showed that NGF mRNA hybridization signal was not found in the submandibular gland of newborn mice, and negative to weak positive expression of NGF in submandibular gland of 8 ~ 10d mice. The signal from the submandibular gland of submandibular gland from 12 to 14 days old mice showed a weak positive signal, located in the cytoplasm of the striatum epithelial cells. Submandibular glandular tubules and granular tubule NGF in situ hybridization signals of 1 month old mice were all positive and located in the cytoplasm around the nucleus. Mice in 2-month-old submandibular glands and granular tubules NGF in situ hybridization signal was strongly positive, mainly in the nucleus of the cytoplasm of the nucleus. The experimental results suggest that the NGF gene in the submandibular gland of male mice begins to be transcribed between 1 and 2 weeks after birth, and the NGF mRNA content increases with age before February
