目的研究miR-146a在结核病患者外周血单个核细胞中的表达水平和临床意义。方法采用Trizonl一步法从结核病患者、结核病潜伏感染者和健康对照外周血单个核细胞提取总RNA,荧光定量PCR测定miR-146a的表达。同时使用荧光定量PCR测定样本中肿瘤坏死因子受体相关因子6、白细胞介素受体相关激酶1和肿瘤坏死因子的表达。结果肺结核患者外周血单核细胞miR-146a表达较结核潜伏感染和健康对照者显著下降,而潜伏感染者miR-146a表达水平(4.49±0.61)与对照组(4.57±0.25)没有差异。同时,miR-146a下游靶基因TRAF6、IRAK-1以及TNF-α表达在活动性肺结核患者中较结核潜伏感染和健康对照者增加。结论与健康对照和潜伏感染者相比,miR-146a在结核病患者外周血单个核细胞中表达显著下降,推测miR-146a有可能成为鉴别活动性结核和结核潜伏感染新的标记物。
Objective To study the expression of miR-146a in peripheral blood mononuclear cells of patients with tuberculosis and its clinical significance. Methods One-step Trizonl method was used to extract total RNA from peripheral blood mononuclear cells of tuberculosis patients, latent tuberculosis patients and healthy controls. The expression of miR-146a was detected by quantitative PCR. At the same time, the expression of tumor necrosis factor receptor-related factor 6, interleukin -receptor associated kinase 1 and tumor necrosis factor-α in samples was determined by fluorescence quantitative PCR. Results The expression of miR-146a in peripheral blood mononuclear cells in patients with tuberculosis was significantly lower than that in latent tuberculosis and healthy controls. However, the expression level of miR-146a in latent infected patients (4.49 ± 0.61) was not different from that in control group (4.57 ± 0.25). At the same time, the expression of TRAF6, IRAK-1 and TNF-α, the downstream targets of miR-146a, were increased in patients with active pulmonary tuberculosis compared with tuberculosis latent infection and healthy controls. Conclusion miR-146a is significantly decreased in peripheral blood mononuclear cells from healthy controls and latent infection patients. It is speculated that miR-146a may be a new marker to identify active tuberculosis and latent tuberculosis infection.
