目的:观察补阳还五汤对局灶性脑缺血损伤模型大鼠微循环血流及相关因子的影响。方法:实验分为假手术组,模型对照组,阿托伐他汀组(10 mg·kg~(-1)),补阳还五汤低、中、高剂量组(5,10,20 g·kg~(-1));采用大脑中动脉栓塞法复制MCAO脑缺血再灌注大鼠模型,参照Longa法进行行为障碍评分;MoorFLPI散斑全帧实时扫描成像系统测定各大鼠尾部微循环血流;ELISA法检测血清TXB2、6-keto-PGF1α水平。结果:各剂量补阳还五汤均能显著降低模型大鼠神经行为学评分(P<0.05),增加尾部微血流量(P<0.05,P<0.01),20 g·kg~(-1)补阳还五汤能显著增加6-keto-PGF1α水平,降低TXB2/6-ketoPGF1α比例。结论:补阳还五汤对MCAO大鼠脑缺血损伤具有改善作用,该作用与改善微循环血流,调节TXB2/6-keto-PGF1α比例有关。
Objective: To observe the effects of Buyang Huanwu Decoction on microcirculation blood flow and related factors in focal cerebral ischemia model rats. Methods: The experiment was divided into sham operation group, model control group, atorvastatin group (10 mg · kg -1), Buyang Huanwu Decoction low, medium and high dose group (5, 10, 20 g · kg ~ (-1)). MCAO cerebral ischemia / reperfusion rat model was reproduced by middle cerebral artery occlusion (MCAO) and behavioral deficit score was assessed according to Longa’s method. MoorFLPI speckle full frame real-time scanning imaging system was used to measure the tail microcirculation Flow; ELISA method to detect serum TXB2, 6-keto-PGF1α levels. Results: Bu Yang Huan Wu decoction could significantly reduce the neurobehavioral score (P <0.05), increase tail micro-blood flow (P <0.05, P <0.01), 20 g · kg -1 Buyang Huanwu Decoction can significantly increase the level of 6-keto-PGF1α, reduce TXB2 / 6-ketoPGF1α ratio. CONCLUSION: Buyang Huanwu decoction can improve cerebral ischemia injury in MCAO rats, which is related to improving microcirculation blood flow and regulating the ratio of TXB2 / 6-keto-PGF1α.
