贵州公路实行“条块结合、以条为主”的管理体制。作为省一级公路管理机构是全省公路行业的主管部门,贵州省公路局负责全省公路的管理和监督工作,直接管养国省干线公路,并作为建设业主承担省内部分高速公路的建设营运任务,同时对农村公路的建设和管养进行行业管理和业务技术指导。贵州省公路局下辖包括9个市、州公路管理局在内的14个企事业单位。主要业务和经营范围包括公路与桥梁的管理、设计、施工、监理以及养路机械制造、物资储备和医疗等。截至2013年底,贵州公路通车里程达到17.3万公里,其中,省公路局直接负责管养1.1万公里。全省公路行业共有职工17885人,其中在职人员8595人,离退休人员9290人。
