目的研究Rho亚家族蛋白(RhoA、B、C)与Rho激酶即ROCK(ROCKⅠ、Ⅱ)在大鼠不同孕期胎盘中的表达水平及其动态变化特点。方法正常受孕的20只雌性大鼠分为孕7、10、13、15、18 d组;各组孕鼠正常饲养,并分别于妊娠第7、10、13、15、18天剖宫取胎盘,测量胎盘湿重及胎盘直径,并记录胎盘个数;采用Western blot及免疫组化法测定胎盘组织中RhoA、B、C与ROCKⅠ、Ⅱ蛋白的表达水平。结果随着孕龄增加,大鼠胎盘的质量和直径都在增加;Westernblot及免疫组化法检测结果显示,RhoA、RhoC的表达水平在孕13、15、18 d组显著高于孕7、10 d组(P<0.01),而RhoB表达水平无明显差异(P>0.05)。ROCKⅠ、ROCKⅡ在孕18 d组表达显著高于孕7、10、13、15 d组(P<0.01)。结论 RhoA、RhoC、ROCKⅠ、ROCKⅡ在大鼠胎盘组织中的表达呈上升趋势,而RhoB则无明显变化;Rho/ROCK信号通路在正常妊娠不同时期的大鼠胎盘组织中发挥重要作用。
Objective To investigate the expression of Rho subfamily protein (RhoA, B, C) and Rho kinase ROCK (ROCK Ⅰ, Ⅱ) in the placenta of different pregnant rats during pregnancy and their dynamic changes. Methods Twenty female rats with normal pregnancy were divided into 7, 10, 13, 15 and 18 d groups. The pregnant rats in each group were reared normally, and the cesarean section was taken on the 7th, 10th, 13th, , Placental wet weight and placental diameter were measured and the number of placenta was recorded. Western blot and immunohistochemistry were used to determine the expression of RhoA, B, C and ROCK Ⅰ, Ⅱ in placenta. Results With the increase of gestational age, the quality and diameter of placenta increased. Western blot and immunohistochemistry showed that the expression levels of RhoA and RhoC were significantly higher than those of pregnant 7, 15 and 18 d d group (P <0.01), while the expression of RhoB had no significant difference (P> 0.05). The expression of ROCK Ⅰ and ROCK Ⅱ in pregnant 18 d group was significantly higher than those in pregnant 7, 10, 13 and 15 d groups (P <0.01). Conclusion The expression of RhoA, RhoC, ROCK Ⅰ and ROCK Ⅱ in rat placenta increased while that of RhoB did not change significantly. The Rho / ROCK signal pathway plays an important role in placental tissue of rats at different stages of normal pregnancy.
