目的探讨梓醇对正常肝细胞系HL-7702细胞胰岛素抵抗的改善作用及其可能机制。方法以MTT法检测梓醇对HL-7702细胞增殖的影响;以高浓度胰岛素及地塞米松作用于HL-7702细胞建立胰岛素抵抗模型。用葡萄糖氧化酶法检测梓醇对胰岛素抵抗HL-7702细胞葡萄糖消耗的影响,Western blot法检测细胞内磷脂酰肌醇-3激酶(PI-3K)蛋白的表达。结果梓醇低、中、高剂量组(5,10,20μmol·L-1)对HL-7702细胞增殖无显著影响,中、高剂量的梓醇可促进胰岛素抵抗HL-7702细胞的葡萄糖消耗,并可增加细胞内PI-3K的表达。结论梓醇可有效改善肝细胞胰岛素抵抗,可能是通过激活PI-3K途径而实现的。
Objective To investigate the effect of catalpol on insulin resistance in normal liver cell line HL-7702 and its possible mechanism. Methods MTT assay was used to determine the effect of catalpol on the proliferation of HL-7702 cells. Insulin resistance was induced by high concentrations of insulin and dexamethasone in HL-7702 cells. The effect of catalpol on glucose depletion in insulin resistance HL-7702 cells was detected by glucose oxidase method. The expression of PI-3K protein in cells was detected by Western blot. Results Catalpol low, medium and high dose groups (5, 10, 20 μmol·L-1) had no significant effect on the proliferation of HL-7702 cells. Catalpol at medium and high doses could promote glucose depletion in HL-7702 cells, And increase intracellular PI-3K expression. Conclusion Catalpol can effectively improve the insulin resistance of hepatocytes, which may be through the activation of PI-3K pathway.
