采用激光拉曼光谱技术研究脱氧核糖核酸 ( DNA)的热变性。实验结果表明 ,DNA热变性可以导致 :1)DNA中脱氧核糖磷酸主链振动减弱 ,包括在 884~ 890 cm-1、1112 cm-1和 1168cm-1处的谱带 ;2 )脱氧核糖 ( d)以及d中 C O的伸缩振动增强 ,在 1436cm-1和 10 72 cm-1处出现了新的谱带 ,在 1466cm-1和 10 30 cm-1处的谱带强度增强 ;3)各碱基的振动频率和相对强度都发生了很大的变化 ,碱基中羰基的伸缩振动显著增强 ,在 1650 cm-1和1670 cm-1处出现了新的谱带 ,在 1710 cm-1处的谱带强度显著增强
Laser Raman spectroscopy was used to study the thermal denaturation of DNA.
