目的 :探讨组织因子(tissue factor,TF)对胃癌SGC7901和MKN45细胞生物学行为的影响。方法 :分别构建反义表达TF和正义表达TF的重组载体pcDNA3.1-antisense TF和pcDNA3.1-sense TF,采用脂质体转染法转入高表达TF的胃癌SGC7901细胞和不表达TF的MKN45细胞中。分别采用MTT法、FCM法及Transwell小室法,观察抑制或过表达TF对胃癌细胞生长、细胞周期分布及侵袭能力的影响。结果 :转染反义表达TF的重组载体后,SGC7901细胞中TF的表达被抑制,与亲本细胞及转染空载体细胞相比,细胞增殖能力明显下降(P<0.05),增殖指数(proliferation index,PI)由亲本细胞的0.357下降至0.286,同时穿过人工基质胶的细胞数目明显减少(P<0.01);而不表达TF的MKN45细胞转染了正义表达TF的重组载体后,与亲本细胞和转染空载体细胞相比,其增殖能力明显增高(P<0.05),PI值由亲本细胞的0.277上升至0.366,同时细胞的侵袭能力明显增加(P<0.01)。结论 :TF可能是一种与胃癌细胞增殖和侵袭相关的调控因子。
Objective: To investigate the effect of tissue factor (TF) on the biological behavior of gastric cancer cell line SGC7901 and MKN45. Methods: Recombinant plasmids pcDNA3.1-antisense TF and pcDNA3.1-sense TF were constructed respectively by antisense TF and sense TF, and transfected into SGC7901 cells with high expression of TF and non-TF expression by lipofection MKN45 cells. The effects of inhibiting or overexpressing TF on the growth, cell cycle distribution and invasion ability of gastric cancer cells were observed by MTT assay, FCM assay and Transwell chamber assay. Results: The expression of TF was inhibited in SGC7901 cells transfected with the recombinant vector with antisense TF. Compared with the parental cells and empty vector transfected cells, the proliferation of cells was significantly decreased (P <0.05). The proliferation index , PI) decreased from 0.357 to 0.286 in the parental cells, while the number of cells passing through the artificial matrigel decreased significantly (P <0.01). However, the MKN45 cells that did not express TF were transfected with the recombinant vector expressing the TF gene directly, Compared with the blank vector transfected cells, the proliferative ability was significantly increased (P <0.05). The PI value increased from 0.277 to 0.366 in the parental cells, and the invasiveness of the cells was significantly increased (P <0.01). Conclusion: TF may be a regulator of proliferation and invasion of gastric cancer cells.
