3种抗氧化剂对Cr6+所致细胞生物学效应的影响

目的:研究维生素C、谷胱甘肽、秦美种猕猴桃汁3种抗氧化剂对铬(Cr)所致细胞毒性、DNA损伤及Cr价态的影响。方法:应用噻唑蓝比色法、单细胞凝胶电泳及原子吸收法研究Cr的细胞毒性、DNA损伤与价态的变化及3种抗氧化剂的干预作用。结果:猕猴桃汁可以有效地拮抗Cr6+引起的细胞毒性,促进Cr6+在细胞外的还原,降低Cr6+的细胞摄入,而维生素C及谷胱甘肽的作用则随Cr染毒剂量的不同而不同。结论:猕猴桃汁对Cr所致细胞生物学效应的拮抗作用明显优于维生素C及谷胱甘肽,可能与其促进Cr6+细胞外还原,阻断Cr6+细胞摄入有关。 Objective: To study the effects of three antioxidants such as Vitamin C, Glutathione and Qin Kiwi juice on the cytotoxicity, DNA damage and Cr valence caused by Cr. Methods: Cytotoxicity, DNA damage and valence of Cr were studied by MTT assay, single cell gel electrophoresis and atomic absorption spectrophotometry. The effects of three antioxidants were also studied. Results: Kiwi juice can effectively antagonize the cytotoxicity induced by Cr6 +, promote the extracellular reduction of Cr6 +, and reduce the cellular uptake of Cr6 +. However, the effect of vitamin C and glutathione varies with the dose of Cr. CONCLUSION: Kiwifruit juice has a significantly better antagonistic effect on cytotoxicity induced by Cr than that of vitamin C and glutathione, which may be related to the promotion of extracellular reduction of Cr6 + and the uptake of Cr6 + cells.
