目的:观察卵巢去势骨质疏松大鼠上颌牙牙本质基质蛋白1(DMP1)的表达变化。方法:选择三月龄健康雌性wistar大鼠40只,在SPF级动物房内适应性饲养一周后,随机分为二组:Sham组(假手术组,仅去除和双侧卵巢等大的脂肪组织)20只;Ovx组(手术组,去除双侧卵巢)20只。实验周期为12 w。术后4 w,8 w,12 w取材,根据实验需要留取腰椎和下颌骨。腰椎用于检测骨密度,下颌骨储存于液氮用于逆转录-聚合酶链反应(RT-PCR)检测不同组别不同时间DMP1的基因水平的表达。结果:术后12 w时,OVX组腰椎骨密度与Sham组相比明显下降,差异有统计学意义(P<0.05)。术后4 w及8 w时,两组之间DMP1的m RNA表达比较无统计学差异(P>0.05),术后12 w时,Ovx组DMP1的m RNA表达显著高于Sham组,差别有统计学意义(p<0.05)。结论:卵巢去势骨质疏松大鼠下颌骨中DMP1 m RNA的表达明显下降,DMP1可能在雌激素缺乏所致骨质疏松的发生过程中具有一定的作用。
Objective: To observe the changes of maxillary dentin matrix protein 1 (DMP1) in ovariectomized rats. Methods: Forty healthy three-month-old female wistar rats were randomly divided into two groups: Sham group (sham operation group, only removing large adipose tissue such as bilateral ovaries and so on) ) 20; Ovx group (surgery group, removing bilateral ovaries) 20. The experimental period is 12 w. After 4 w, 8 w and 12 w, the lumbar and mandible were collected according to the experiment. The lumbar spine was used to detect the bone mineral density. The mandible was stored in liquid nitrogen for reverse transcription-polymerase chain reaction (RT-PCR) to detect the expression of DMP1 at different time points. Results: At 12 weeks after operation, the lumbar BMD of OVX group was significantly lower than that of Sham group (P <0.05). At 4 weeks and 8 weeks after operation, there was no significant difference in the mRNA expression of DMP1 between the two groups (P> 0.05). At 12 weeks after operation, the mRNA expression of DMP1 in Ovx group was significantly higher than that in Sham group Statistical significance (p <0.05). Conclusion: The expression of DMP1 mRNA in the mandible of ovariectomized rats with osteoporosis significantly decreased. DMP1 may play a role in the pathogenesis of osteoporosis induced by estrogen deficiency.
