药物的能穿透皮肤对疗效有重要意义。过去認为药物的进入皮肤都经毛孔,其直接通过表皮者微不足道,现在实验证明药物可以穿透表皮。药物的脂溶性是穿透皮肤的主要因素,而毛孔中分泌的脂肪性物质亦有促进脂溶性药物的穿透作用。皮肤有很多汗腺,水溶性药物可通过汗腺进入皮肤。菸酸,水杨酸等经制成甲酯后由于其油溶性增加,极易透过皮肤。
