目的分析化脓性眼内炎患者感染病原菌及其对蛋白激酶B(AKT)通路蛋白水平的影响研究,为临床相关治疗提供借鉴。方法对医院2011年4月-2014年4月诊治的110例化脓性眼内炎患者进行研究,应用VITEK-2Compact全自动鉴定仪对病原菌进行分型,酶联免疫吸附测定法测定感染后患者血清中AKT、磷脂酰肌醇3-激酶(PI3K)及脂肪酸合成酶(Fas)蛋白水平的变化,同时分析患者血清中氧化/抗氧化酶的变化,以医院同期80名健康体检者为对照组,以80例未感染眼内炎患者为未感染组。结果患者中分离病原菌110株,革兰阳性菌66株占60.0%,革兰阴性菌44株占40.0%;与对照组和未感染组患者相比,感染组患者血清中AKT、PI3K及Fas但水平明显升高,丙二醛(MDA)水平升高,而超氧化物歧化酶(SOD)和过氧化氢酶(CAT)水平明显降低,且差异均有统计学意义(P<0.01)。结论化脓性眼内炎感染病原菌以革兰阳性菌为主,且能上调血清中AKT、PI3K及Fas和MDA、下调SOD及CAT水平。
Objective To analyze the pathogenic bacteria infected with purulent endophthalmitis and its effect on the protein level of protein kinase B (AKT), so as to provide reference for clinical treatment. Methods A total of 110 cases of purulent endophthalmitis treated in our hospital from April 2011 to April 2014 were studied. Pathogenic bacteria were classified by VITEK-2 Compact automatic analyzer. Serum samples were determined by enzyme-linked immunosorbent assay AKT, phosphoinositide 3-kinase (PI3K) and fatty acid synthase (Fas) protein levels in patients with serum levels of oxidative / anti-oxidant changes in the same period to the hospital 80 healthy people as control group, Eighty non-infected endophthalmitis patients were uninfected. Results Among the patients, 110 strains of pathogens were isolated, 66.0% of Gram-positive bacteria and 44.0% of Gram-negative bacteria. Compared with control group and non-infected group, AKT, PI3K and Fas in serum of infected patients (P <0.01). The level of malondialdehyde (MDA) increased significantly while the levels of superoxide dismutase (SOD) and catalase (CAT) decreased significantly (P <0.01). Conclusions The pathogens of purulent endophthalmitis are mainly Gram-positive bacteria, and can up-regulate AKT, PI3K, Fas and MDA in serum, and down-regulate the levels of SOD and CAT.
