目的:观察清热解毒、活血化瘀方对胰腺癌移植瘤的抗肿瘤作用及其NF-κB表达的影响,探讨该组方的抗肿瘤机制。方法:40只胰腺癌移植瘤裸鼠分为对照组、低、中、高剂量组,观察各组对移植瘤瘤重的影响,并计算抑瘤率,并用RT-PCR方法测定各组NF-κB的相对含量,比较各组对NF-κB mRNA表达的影响。结果:①清热解毒、活血化瘀方低、中、高剂量的抑瘤率分别为36.29%、45.72%、57.53%,呈剂量依赖性,与对照组比较差异显著(P<0.01);②清热解毒、活血化瘀方低、中、高剂量组NF-κBmRNA相对表达分别为1.64±0.39、1.14±0.41、0.62±0.38,中、高剂量组与对照组比较差异显著(P<0.01),而低剂量组与对照组比较差异虽无统计学意义(P>0.05),但与对照组相比较,NF-κB表达具有下降的趋势。结论:清热解毒、活血化瘀方对胰腺癌裸鼠移植瘤具有一定的抑制肿瘤生长作用,下调NF-κB基因的表达是其抗肿瘤机制的主要途径之一。
Objective: To observe the anti-tumor effect of Qingre Jiedu, Huoxue Huayu Recipe on pancreatic cancer xenografts and its effect on the expression of NF-κB, and to explore the anti-tumor mechanism of this recipe. Methods: Forty pancreatic cancer xenografts in nude mice were divided into control, low, middle and high dose groups. The effects of each group on tumor weight were observed. The inhibition rate of tumor was calculated. The expression of NF- κB relative content of each group compared the impact of NF-κB mRNA expression. Results: ① The tumor inhibition rates of Qingrejiedu, Huoxuehuayu Recipe were 36.29%, 45.72% and 57.53%, respectively, in a dose-dependent manner, with significant difference compared with the control group (P <0.01); ② Heat-clearing The relative expression of NF-κB mRNA in low, medium and high dose groups of detoxification and promoting blood circulation to remove blood stasis were 1.64 ± 0.39,1.14 ± 0.41 and 0.62 ± 0.38, respectively. The middle and high dose groups showed significant difference compared with the control group (P <0.01) The difference between the low-dose group and the control group was not statistically significant (P> 0.05), but compared with the control group, the expression of NF-κB had a decreasing trend. Conclusion: Qingrejiedu and Huoxuehuayu Recipe can inhibit the growth of pancreatic cancer xenografts in nude mice. Down-regulation of NF-κB gene expression is one of the main ways of antitumor mechanism.
