以5份优良玉米自交系为试材,采用农杆菌侵染玉米萌动胚的方法转化蓝铜蛋白类似基因BcBCP1,同时优化遗传转化体系。结果表明,菌液浓度为OD600=0.8、侵染24h、共培养3d时,遗传转化率分别达到最高;不同基因型的遗传转化率存在差异,其中丹598转化率最高,达到6.67%;丹599的遗传转化率最低,为0.56%,5个不同基因型的平均遗传转化率为2.13%。共获得PCR阳性植株43株,其中28株结实。
Five elite maize inbred lines were used as experimental materials, Agrobacterium tumefaciens was used to transform BmBCP1, and the genetic transformation system was optimized. The results showed that the genetic transformation rates reached the highest when OD600 = 0.8, infection 24h and co-culture for 3d, respectively. The genetic transformation rates of different genotypes were different, of which Dan 598 had the highest conversion rate of 6.67% and Dan 599 Had the lowest genetic conversion rate of 0.56%. The average genetic conversion rate of 5 different genotypes was 2.13%. A total of 43 PCR-positive plants were obtained, of which 28 were strong.
