目的 研究SARS病毒合成肽疫苗。方法 根据目前已知SARS病毒的基因组序列及其编码的蛋白质 ,选择了SARS病毒编码的spike蛋白抗原表位蛋白质序列 ,运用计算机抗原表位预测技术 ,筛选抗原表位 ,以合成肽抗原表位。通过对SARS病人血清进行筛选确定与血清高交叉反应的 3个短肽 ,再合成多重抗原肽 (multipleantigenicpeptide ,MAP) ,与KLH偶联后免疫小鼠。结果 筛选的 3条与SARS病人血清反应强的肽 ,免疫小鼠均产生了高效价的抗体。结论 以合成肽为免疫原研究SARS病毒肽疫苗有着广阔前景。
Objective To study SARS virus synthetic peptide vaccine. Methods According to the genome sequence of the known SARS virus and its encoded protein, the protein sequence of spike protein epitope encoded by SARS virus was selected. The antigenic epitopes of the spike protein were screened by using the computer epitope prediction technique to synthesize peptide epitopes. Sera of SARS patients were screened to identify three short peptides that highly cross-reacted with serum. Multipleantigenic peptide (MAP) was synthesized and conjugated to KLH to immunize mice. Results Three peptides that were highly reactive to SARS patients were screened and the immunized mice produced high titer antibody. Conclusion The synthetic peptide as the immunogen to study the SARS virus peptide vaccine has broad prospects.
