目的观察趋化因子CCL19在结直肠癌组织中的表达,探讨其对结直肠癌细胞的增殖、迁移、侵袭能力的影响。方法收集确诊为结直肠恶性肿瘤并手术切除患者(n=85)的肿瘤组织及癌旁正常组织,运用实时定量PCR和组织微阵列免疫组化技术检测CCL19的表达水平;以实时定量PCR、蛋白质印迹技术筛选高表达CCL19受体CCR7的人结直肠癌细胞株SW620细胞作为研究对象,并给予重组人CCL19(rh-CCL19)刺激,通过细胞增殖实验、划痕实验、Transwell实验来检测SW620细胞的增殖、迁移和侵袭能力的变化。结果结直肠癌组织中的CCL19表达要明显低于癌旁正常组织(P<0.05),且CCL19表达量与肿瘤大小和浸润深度有关(P<0.01)。在CCR7高表达的SW620细胞中,加入rh-CCL19后,其细胞增殖、迁移及侵袭能力下降(P<0.05)。结论 CCL19在结直肠癌组织中的表达量低于癌旁正常组织,且与肿瘤大小和浸润深度有关;CCL19可抑制人结直肠癌细胞株SW620细胞增殖、迁移和侵袭能力,提示CCL19具有抑制结直肠癌的作用。
Objective To observe the expression of chemokine CCL19 in colorectal cancer and explore its effect on the proliferation, migration and invasion of colorectal cancer cells. Methods Tumor tissues and adjacent normal tissues were collected from patients diagnosed with colorectal malignant tumors (n = 85). The expression of CCL19 was detected by real-time quantitative PCR and tissue microarray immunohistochemistry. Real-time PCR, Western blotting was used to screen human colorectal cancer cell line SW620, which is highly expressed CCR19 receptor CCR7, and stimulated with recombinant human CCL19 (rh-CCL19). The cell proliferation assay, scratch assay and Transwell assay were used to detect SW620 cells Changes in proliferation, migration and invasiveness. Results The expression of CCL19 in colorectal cancer tissues was significantly lower than that in adjacent normal tissues (P <0.05). The expression of CCL19 was correlated with tumor size and depth of invasion (P <0.01). In CCR7-overexpressing SW620 cells, the cell proliferation, migration and invasion decreased after addition of rh-CCL19 (P <0.05). Conclusion The expression of CCL19 in colorectal cancer tissues is lower than that in adjacent normal tissues, which is related to tumor size and depth of invasion. CCL19 can inhibit the proliferation, migration and invasion of human colorectal cancer cell line SW620, suggesting that CCL19 has an inhibitory effect The role of rectal cancer.
