目的探讨程序性细胞凋亡因子4(PDCD4)与转化生长因子β1(TGF-β1)在急性髓系白血病(AML)患者骨髓中的表达及其相关关系。方法采用RT-PCR、ELISA和Western blotting三种方法分别从基因水平和蛋白水平检测30例初诊急性髓系白血病患者(AML组),30例急性髓系白血病完全缓解患者(AML-CR组),20例缺铁性贫血患者(对照组)骨髓单个核细胞中PDCD4、TGF-β1的表达情况,并分析相互间的相关性。结果 PDCD4、TGF-β1的RT-PCR结果均显示,AML组mRNA的表达量明显低于对照组(P<0.05),AML-CR组表达量虽明显高于AML组,但差异没有统计学意义(P>0.05)。ELISA结果显示,AML组TGF-β1的蛋白表达量明显低于对照组和AML-CR组(P<0.05)。Western blotting结果显示,AML组PDCD4蛋白表达明显低于对照组和AML-CR组(P<0.05)。PDCD4、TGF-β1的表达呈正相关关系(P<0.05)。结论 PDCD4、TGF-β1在初诊AML中低表达,PDCD4、TGF-β1在白血病的发生发展中具有相关关系。
Objective To investigate the expression of programmed cell death factor 4 (PDCD4) and transforming growth factor-β1 (TGF-β1) in the bone marrow of patients with acute myeloid leukemia (AML) and its relationship. Methods Thirty newborns with acute myeloid leukemia (AML group), 30 acute leukemia patients with complete remission (AML-CR group) and 30 normal controls were enrolled in this study. 20 cases of iron deficiency anemia (control group) bone marrow mononuclear cells PDCD4, TGF-β1 expression, and analyze the correlation between each other. Results The RT-PCR results of PDCD4 and TGF-β1 showed that the expression of mRNA in AML group was significantly lower than that in control group (P <0.05), while the expression of AML-CR group was significantly higher than that in AML group, but the difference was not statistically significant (P> 0.05). The results of ELISA showed that the protein expression of TGF-β1 in AML group was significantly lower than that in control group and AML-CR group (P <0.05). Western blotting showed that PDCD4 protein expression in AML group was significantly lower than that in control group and AML-CR group (P <0.05). PDCD4, TGF-β1 expression was positively correlated (P <0.05). Conclusion The expression of PDCD4 and TGF-β1 is low in newly diagnosed AML, and the expression of PDCD4 and TGF-β1 is correlated with the occurrence and development of leukemia.
