目的:观察50℃时不同热疗时间对CT-26细胞的损伤作用。方法:将CT-26细胞在50℃水浴中加热至各时间点(5、10、15、20和30min),后采用不同的方法分别观察不同时间热疗后的细胞形态、细胞生存率和细胞生长周期及凋亡情况。结果:细胞生存率由5min时的72.3%降至30min时的1.7%(MTT法),由72.9%降至1.4%(台盼蓝染色排除法)。5min时细胞凋亡率为(28.047±0.675)%,在各时间组中最高,P<0.05。结论:50℃热疗,<5min以诱导细胞凋亡为主;10min时不仅引起细胞坏死,还可诱导细胞凋亡;>15min以细胞坏死为主。
Objective: To observe the effect of different hyperthermia time on CT-26 cell injury at 50 ℃. Methods: CT-26 cells were heated in water bath at 50 ℃ for 5, 10, 15, 20 and 30 min. Different methods were used to observe the cell morphology, cell viability and cell survival rate Growth cycle and apoptosis. Results: The cell viability decreased from 72.3% at 5 min to 1.7% at 30 min (MTT method) from 72.9% to 1.4% (trypan blue exclusion method). The apoptosis rate at 5 min was (28.047 ± 0.675)%, which was highest in each time group (P <0.05). Conclusion: 50 ℃ hyperthermia, <5min to induce apoptosis; 10min not only cause cell necrosis, but also induce apoptosis;> 15min to cell necrosis.
