目的:明确孤儿受体—雌激素受体相关受体(estrogen receptor-related recep-tor,ERRs)各亚型以及雌激素受体亚型在卵巢癌中mRNA的表达模式。方法:采用Light-Cycler-PCR定量检测40例卵巢癌标本以及15例正常卵巢组织中各种ERRs亚型和ERs亚型mRNA的表达丰度。采用ELISA方法分析各样本血清CA-125水平。结果:卵巢癌组织中ERRαmRNA表达的丰度和阳性表达率显著高于正常卵巢组织(P=0.04和P=0.02),ERRβ在卵巢癌组织和正常卵巢组织中的RNA表达的丰度和阳性表达率差异无统计学意义(P>0.05);ERRγmRNA在正常卵巢以及卵巢癌组织中表达的丰度差异无统计学意义(P>0.05),但卵巢癌组织中ERRγ的阳性表达率显著增高(P=0.045)。ERRs家族和ERs家族在卵巢癌中有很高的共同表达率,ERα/ERRα的比率在卵巢癌组织中明显下降(P=0.0412)。结论:ERRs与ERs在卵巢癌中有共同表达,两个家族间的相互作用可能是卵巢癌复杂的肿瘤内分泌生物学行为的分子基础。
OBJECTIVE: To clarify the expression patterns of orphan receptor-related recep-tor (ERRs) subtypes and estrogen receptor subtypes in ovarian cancer. Methods: The expression abundance of various ERRs subtypes and ERs subtypes in 40 cases of ovarian cancer and 15 cases of normal ovarian tissue were detected by Light-Cycler-PCR. Serum CA-125 levels were analyzed by ELISA. Results: The abundance and positive expression of ERRα mRNA in ovarian cancer tissues were significantly higher than those in normal ovarian tissues (P = 0.04 and P = 0.02). The abundance and positive expression of ERRβ in ovarian cancer tissues and normal ovarian tissues (P> 0.05). There was no significant difference in the expression of ERRγ mRNA between normal ovary and ovarian cancer (P> 0.05), but the positive rate of ERRγ in ovarian cancer was significantly higher than that of normal = 0.045). The ERRs family and the ERs family have a high common expression rate in ovarian cancer, and the ratio of ERα / ERRα is significantly decreased in ovarian cancer (P = 0.0412). Conclusions: ERRs and ERs are co-expressed in ovarian cancer. The interaction between the two families may be the molecular basis of the complex tumor endocrine biological behavior of ovarian cancer.
