目的:观察橙黄决明素对实验性高脂血症大鼠血脂水平的影响。方法:以控制LDLR基因表达的SRE为靶位点,构建LDLR启动子荧光素酶报告基因质粒,将此质粒转染人肝癌细胞系HepG2构建成LDLR报告基因模型,利用该模型研究橙黄决明素的降血脂活性;采用脂肪乳剂致大鼠高脂血症模型,测定各组动物的血清总胆固醇(TC)、甘油三脂(TG)和低密度脂蛋白胆固醇(LDL-C)。结果:橙黄决明素对低密度脂蛋白受体基因转录水平有增强作用,橙黄决明素能显著降低高脂血症大鼠的TC、LDL-C和TG。结论:橙黄决明素具有降血脂作用。
OBJECTIVE: To observe the effect of orange scutellarin on blood lipid levels in experimental hyperlipidemic rats. Methods: The LDLR promoter luciferase reporter gene plasmid was constructed with SRE as the target site to control the LDLR gene expression. The LDLR reporter gene model was constructed by transfecting this hepatocellular carcinoma cell line HepG2 into LDLR reporter gene model. The lipid-lowering activity; rat model of hyperlipidemia induced by fat emulsion, determination of serum total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) in each group of animals. RESULTS: Orange yellow desmosin enhanced the transcription of low-density lipoprotein receptor gene, and orange yellow desmin significantly reduced TC, LDL-C and TG in hyperlipidemic rats. Conclusion: Orange yellow desmosin has hypolipidemic effect.
