胃炎饮方对胃溃疡大鼠表皮生长因子和转化生长因子β1表达的影响

目的从调节表皮生长因子(EGF)和转化生长因子β1(TGF-β1)表达的角度探讨胃炎饮方促进胃溃疡愈合的分子机制。方法 50只Wistar大鼠随机抽取8只作为假手术组,其余42只采用冰醋酸腐蚀法建立胃溃疡大鼠模型。将造模成功的32只分为模型组、胃高组、胃低组、对照组,每组8只。胃高组、胃低组分别灌服生药浓度为2.14、1.07 g/ml胃炎饮煎剂10 ml/kg,对照组灌服浓度为0.368 mg/ml的奥美拉唑液10 ml/kg。各给药组从造模术后第3天开始灌胃,假手术组、模型组灌胃等量生理盐水,每日1次,共14天。双抗体夹心ABC-ELISA法检测各组大鼠EGF、TGF-β1水平,免疫组化方法检测各组大鼠EGF、TGF-β1表达,RT-PCR方法检测各组大鼠EGF mRNA、TGF-β1mRNA的表达。结果与假手术组比较,模型组大鼠胃组织EGF、TGF-β1水平显著升高,平均光度值显著升高,而平均灰度值显著下降,EGF mRNA、TGF-β1mRNA表达显著升高(P<0.05或P<0.01);与模型组比较,各治疗组EGF、TGF-β1水平及平均光度值显著升高,平均灰度值显著下降,EGF mRNA、TGF-β1mRNA表达显著升高,溃疡面积均明显缩小(P<0.05或P<0.01);与对照组和胃低组比较,胃高组EGF、TGF-β1水平及平均光度值显著升高(P<0.05或P<0.01),EGF mRNA、TGF-β1mRNA表达及溃疡面积差异具有统计学意义(P<0.05或P<0.01)。结论胃炎饮可促进胃溃疡大鼠胃组织EGF和TGF-β1表达,加强胃黏膜修复,可能是提高溃疡愈合的可能机制。 Objective To investigate the molecular mechanism of Weiyan Decoction in promoting the healing of gastric ulcer from the perspective of regulating the expression of epidermal growth factor (EGF) and transforming growth factor-β1 (TGF-β1). Methods Fifty Wistar rats were randomly divided into sham operation group and the remaining 42 animals were treated with acetic acid acetic acid method to establish gastric ulcer rat model. 32 successful models were divided into model group, high gastric group, low gastric group, control group, 8 in each group. Gastric high group, low gastric group were dosed with crude drug concentration of 2.14,1.07 g / ml gastritis decoction 10 ml / kg, the control group orally fed omeprazole concentration of 0.368 mg / ml 10 ml / kg. Each administration group began to gavage on the 3rd day after the modeling operation, and the sham-operation group and model group were given the same amount of normal saline once a day for 14 days. The levels of EGF and TGF-β1 were detected by double antibody sandwich ABC-ELISA in each group. The expression of EGF and TGF-β1 in each group were detected by immunohistochemical method. The expressions of EGF mRNA, TGF-β1 mRNA expression. Results Compared with the sham operation group, the levels of EGF and TGF-β1 in the gastric tissue of model group were significantly increased, the average photometric value was significantly increased, while the average gray value was significantly decreased, while the expression of EGF mRNA and TGF-β1 mRNA was significantly increased (P <0.05 or P <0.01). Compared with the model group, the levels of EGF, TGF-β1 and the average photometric value of each treatment group were significantly increased, the average gray value was significantly decreased, the expression of EGF mRNA and TGF-β1 mRNA were significantly increased, the ulcer area (P <0.05 or P <0.01). Compared with the control group and the gastric low group, the levels of EGF, TGF-β1 and the average photometric value in the gastric hyperglycemia group were significantly increased (P <0.05 or P <0.01) , TGF-β1mRNA expression and ulcer area difference was statistically significant (P <0.05 or P <0.01). Conclusions Weiyan Decoction can promote the expression of EGF and TGF-β1 in stomach tissue of gastric ulcer rats, and strengthen gastric mucosa repair may be a possible mechanism to improve the healing of ulcer.
