目的:研究泛肽相关新基因编码产物的特性和在鼻咽癌细胞内定位与表达。方法:利用生物信息学分析编码蛋白的一般性质并预测其定位,构建增强型绿色荧光蛋白(enhance green fluorescent prote in,EGFP)与UBAP1融合基因的真核表达载体pEGFP-C2-UBAP1,通过脂质体介导转染人鼻咽癌细胞系HNE1,荧光显微镜观察UBAP1基因编码蛋白在HNE1活细胞内定位。结果:融合蛋白可在HNE1细胞中高表达,主要定位在细胞核,有较为明显的核膜聚集现象。结论:UBAP1基因编码产物在HNE1中的表达差异可能是NPC发生的原因之一。
OBJECTIVE: To study the characteristics of the novel gene encoding the ubiquitin and its localization and expression in nasopharyngeal carcinoma cells. METHODS: The bioinformatics method was used to analyze the general properties of the encoded protein and to predict its location. The eukaryotic expression vector pEGFP-C2-UBAP1 with enhanced green fluorescent protein (EGFP) and UBAP1 fusion gene HNE1 was transfected into human nasopharyngeal carcinoma cell line HUVECs. Fluorescence microscopy was used to observe the localization of UBAP1 gene in HNE1 live cells. Results: The fusion protein was highly expressed in HNE1 cells, mainly located in the nucleus, with a more obvious nuclear membrane aggregation phenomenon. Conclusion: The difference in the expression of UBAP1 gene in HNE1 may be one of the causes of NPC.
