用造血祖细胞体外培养、造血生长因子检测和流式细胞术等方法,研究当归多糖(AP)诱导L-细胞(成纤维细胞株)产生造血生长因子及其对血细胞发生的调节作用。结果表明,L-细胞条件培养液(LcCM)对造血祖细胞(CFU-GM、BFU-E、CFU-E)的体外增殖呈双向调节作用;10%LcCM能促进骨髓造血细胞进入增殖活跃的S+M/G2期;经AP诱导制备的LcCM在有或无外源性造血生长因子(如Epo,BPA,GM-CSF)存在时均对造血祖细胞的克隆增殖显示较高刺激活性;AP或IL-1与造血生长因子间似无协同作用。本研究提示AP可能通过诱导造血微环境的成纤维细胞分泌某些造血生长因子,从而促进造血祖细胞增殖分化,这或许是当归"补血"的生物学机理之一。
Hematopoietic progenitor cell culture, hematopoietic growth factor assay and flow cytometry were used to study the effect of angelica polysaccharide (AP) on hematopoietic growth factors induced by L-cells (fibroblasts) and their regulation of hematopoiesis. The results showed that L-cell conditioned medium (LcCM) showed biphasic regulation of hematopoietic progenitor cells (CFU-GM, BFU-E, CFU-E) in vitro; 10% LcCM promoted bone marrow hematopoietic cells to proliferate actively S+M /G2 phase; LcCM induced by AP showed high stimulatory activity on the proliferation of hematopoietic progenitor cells in the presence or absence of exogenous hematopoietic growth factors (such as Epo, BPA, GM-CSF); AP or IL- 1 There is no synergy with hematopoietic growth factors. This study suggests that AP may promote certain hematopoietic growth factors by inducing hematopoietic microenvironmental fibroblasts, thereby promoting the proliferation and differentiation of hematopoietic progenitor cells, which may be one of the biological mechanisms of Angelica sinensis “blood”.
