目的应用Stereo Investigator 8.0测量小鼠脑切片厚度的信度研究。方法雄性4 mC57BL/6J小鼠14只,随机分40μm、60μm组,制备脑冰冻切片。应用Stereo Investigator 8.0系统测量切片厚度,测量分为单人重复测量、3人测量;分析测量的一致性。结果 40μm组单人重复测量一致性好,3人测量一致性存在轻度可疑;60μm组单人重复测量和3人分别测量一致性均好。结论应用Stereo Investigator 8.0体视学系统,单人重复测量40μm及60μm冰冻切片染色后厚度结果一致稳定;不同人测量一致性略显不足。
Objective To study the reliability of measuring the thickness of brain slices of mice by using Stereo Investigator 8.0. Methods Fourteen male 4 mC57BL / 6J mice were randomly divided into 40 μm and 60 μm groups to prepare brain frozen sections. Slice thickness was measured using the Stereo Investigator 8.0 system. The measurements were divided into single-person repeat measurements, 3-person measurements, and analysis of the agreement. Results The consistency of single-person repeated measurement was good in 40μm group, slight suspiciousness was observed in 3-person measurement. The single-person repeated measurement in 60μm group and the consistency in 3-person measurement were all good. Conclusion Stereo Investigator 8.0 stereo vision system, the single repeat measurements of 40μm and 60μm frozen sections stained the thickness of the results were consistent; slightly different from the consistency of different measurements.
