水胺硫磷对鲫鱼肝微粒体酶CYP450和CYP1A1活性的影响

[目的]观察鲫鱼静态暴露水胺硫磷对肝微粒体CYP450酶系活性的影响,探讨水胺硫磷污染水体的生物学效应。[方法]将鲫鱼分为5.0、10.0、15.0、20.0 mg/L 4个染毒组和1个对照组,进行水胺硫磷静态暴露染毒,在染毒24、48、72、96 h时动态监测鲫鱼肝微粒体CYP450酶及其亚酶CYP1A1的活性,同时用气相色谱法检测试验液中水胺硫磷水平,分析与肝CYP450酶及CYP1A1活性的相关性。[结果]当染毒24 h时,与对照组相比,各染毒组肝微粒体总CYP450酶活性无显著变化,20.0 mg/L组CYP1A1酶活性高于对照组(P<0.05);染毒48 h后所有染毒组总CYP450酶及CYP1A1酶活性较对照组均升高(P<0.01);当染毒96 h时,20.0 mg/L组总CYP450及CYP1A1酶活性分别是对照组的1.29倍和1.54倍。染毒24 h时试验液中水胺硫磷残留量与总CYP450酶及CYP1A1活性无相关关系(P>0.05),但在染毒48 h后就出现了显著的正相关关系,相关系数r均大于0.9(均P<0.05)。[结论]水胺硫磷暴露会引起鲫鱼肝CYP450酶及其亚酶CYP1A1的活性增加,且在暴露48 h以后水体中水胺硫磷的含量和CYP450酶及其亚酶CYP1A1的活性呈正相关关系。 [Objective] The research aimed to observe the effect of static exposure of methamidophil on the microsomal CYP450 enzyme activity of crucian and investigate the biological effect of methamidophos on the water body. [Method] The crucian carp was divided into 4 groups of 5.0, 10.0, 15.0 and 20.0 mg / L and one control group, The activity of CYP450 and CYP1A1 in liver microsomes of Carassius auratus were dynamically monitored. Meanwhile, the content of amilion in test liquid was detected by gas chromatography, and the correlation between CYP450 activity and CYP1A1 activity was analyzed. [Result] Compared with the control group, the total CYP450 enzyme activity of liver microsomes in each exposure group had no significant change when exposed to 24 h. The activity of CYP1A1 in 20.0 mg / L group was higher than that of the control group (P <0.05) After 48 h, the total CYP450 and CYP1A1 activities of all the exposed groups were higher than those of the control group (P <0.01). When exposed to 96 h, the total CYP450 and CYP1A1 activities of 20.0 mg / L group were respectively 1.29 times and 1.54 times. There was no significant correlation between the residual amount of amilion in the test solution and total CYP450 and CYP1A1 activity (P> 0.05) at 24 h after exposure, but a significant positive correlation was observed after 48 h Greater than 0.9 (all P <0.05). [Conclusion] Exposure of amisulphidine could increase the activity of hepatic CYP450 enzyme and its enzyme CYP1A1 in Carassius auratus, and there was a positive correlation between the content of chlorisothion and the activity of CYP450 and its enzyme CYP1A1 after 48 h of exposure .
