目的观察人参皂甙Rg1对糖尿病肾病大鼠基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)蛋白表达的影响。方法采用链脲佐菌素(65mg/kg)腹腔注射建立糖尿病大鼠模型。将40只雄性SD大鼠随机分为4组:正常对照组、糖尿病模型组、人参皂甙Rg1治疗组、厄贝沙坦(ARB)治疗组。8周后检测各种生化指标。HE染色观察肾组织的病理变化。采用免疫组织化学检测肾脏组织MMP-9蛋白水平的表达。结果与对照组相比,模型组大鼠肾小球及肾小管中MMP-9的表达明显减弱。Rg1及ARB治疗组较模型组肾小球及肾小管中MMP-9蛋白的表达明显增加(P<0.05),两治疗组之间较无明显差异。结论人参皂甙Rg1能增加MMP-9的水平,减少糖尿病肾病大鼠肾组织细胞外基质的集聚,减轻肾脏的纤维化。
Objective To observe the effect of ginsenoside Rg1 on the expression of matrix metalloproteinase-9 (MMP-9) in diabetic nephropathy rats. Methods Diabetic rats were induced by intraperitoneal injection of streptozotocin (65mg / kg). Forty male SD rats were randomly divided into 4 groups: normal control group, diabetic model group, ginsenoside Rg1 treatment group and irbesartan (ARB) treatment group. After 8 weeks to detect a variety of biochemical indicators. The pathological changes of kidney were observed by HE staining. Immunohistochemistry was used to detect the expression of MMP-9 in kidney. Results Compared with the control group, the expression of MMP-9 in glomeruli and tubules in model group was significantly decreased. Compared with model group, the expression of MMP-9 protein in glomeruli and tubules in Rg1 and ARB treatment groups was significantly increased (P <0.05), and there was no significant difference between the two treatment groups. Conclusion Ginsenoside Rg1 can increase the level of MMP-9, reduce the accumulation of extracellular matrix in the kidney of diabetic nephropathy rats and reduce the renal fibrosis.
