目的观察β-榄香烯对血管内皮细胞增殖,细胞周期以及成血管能力的影响,探讨β-榄香烯对肿瘤血管生成的抑制作用。方法人脐静脉内皮细胞ECV-304体外培养,β-榄香烯干预;四甲基偶氮唑盐(MTT)法检测β-榄香烯对细胞增殖的抑制作用;流式细胞仪检测细胞周期;Matrigel胶检测细胞成血管能力。结果ECV-304细胞经过β-榄香烯干预,其存活率明显降低,并呈计量依赖性。细胞周期检测结果显示,β-榄香烯干预后,G1期细胞增多,进入S期及G2期细胞明显减少,细胞周期被阻滞于G1期;接种于Matrigel胶的ECV-304细胞经β-榄香烯干预后,形成管腔数目减少,成血管能力明显降低。结论β-榄香烯可直接干扰血管内皮细胞的增殖及细胞周期,阻滞细胞从G1期进入S期及G2期,并降低其成血管能力。
Objective To observe the effect of β-elemene on proliferation, cell cycle and angiogenesis of vascular endothelial cells (ECs), and to investigate the inhibitory effect of β-elemene on tumor angiogenesis. Methods Human umbilical vein endothelial cells (ECV-304) were cultured in vitro and treated with β-elemene. The inhibitory effect of β-elemene on cell proliferation was detected by MTT assay. The cell cycle was detected by flow cytometry Matrigel was used to detect the angiogenic ability of the cells. Results After ECV-304 cells were treated with β-elemene, the survival rate of ECV-304 cells was significantly reduced and in a dose-dependent manner. Cell cycle analysis showed that β-elemene increased the number of cells in G1 phase, reduced the number of cells in S phase and G2 phase, and the cell cycle was arrested in G1 phase. ECV-304 cells seeded in Matrigel showed no significant difference After the elemene intervention, the number of the formation of the lumen is reduced and the ability of angiogenesis is obviously reduced. Conclusion β-elemene directly interferes with the proliferation and cell cycle of vascular endothelial cells, arrests cells from G1 phase into S phase and G2 phase, and decreases their ability to form blood vessels.
