目的用乙烯硫脲诱发妊娠大白鼠产生泄殖腔畸形动物模型,观察其肛周肌肉病理改变。方法取泄殖腔畸形鼠仔,正中矢状面连续切片,用LuzexF图像分析仪测内、外括约肌及耻骨直肠肌面积。用肌球蛋白ATP酶染色,观察鼠仔盆底横纹肌复合体中Ⅰ、Ⅱ型肌纤维发育情况。结果各种泄殖腔畸形内括约肌面积较正常鼠增大,后壁更明显(P<0.05);肛门外括约肌面积较正常鼠明显小;低位泄殖腔畸形耻骨直肠肌面积较正常鼠增大,而中高位其面积较正常鼠小。泄殖腔畸形鼠仔盆底横纹肌复合体中Ⅰ型肌纤维占比例明显减少(P<0.05)。结论泄殖腔畸形动物模型病理改变与人类极相似,为研究该畸形病理提供参考指标
Objective To induce the cloaca model in pregnant rats induced by ethylenethiourea and observe the pathological changes of perianal muscles. Methods The cloacal malformation rat, median sagittal continuous section, with Luzex F image analyzer measured internal and external sphincter and puborectalis muscle area. Myosin ATPase staining was used to observe the development of type I and type II myofibers in the rhabdomyos spine complex. Results The sphincter area in all kinds of cloacal malformations was larger than that in normal rats, and the posterior wall was more obvious (P <0.05). The area of anal external sphincter was significantly smaller than that in normal mice. The area of puborectalis muscle in lower cloaca malformations was larger than that in normal rats Its area is higher than normal mice. The proportion of type Ⅰ myofibers in the pelvic floor striated muscle complex of the cloacal malformations was significantly decreased (P <0.05). Conclusion The pathological changes of cloacal malformation animal model are very similar to those of humans, providing a reference index for studying the deformity pathology
