目的:探讨水通道蛋白5(AQP5)在人宫颈癌中的表达情况及其临床意义。方法:分别通过qRT-PCR和Western blotting方法检测宫颈癌及癌旁组织中AQP5 mRNA和蛋白的表达情况,并分析AQP5 mRNA的表达与临床病理特点的关系。采用ROC曲线分析癌症患者组织中AQP5 mRNA的诊断价值。结果:AQP5 mRNA和蛋白水平在宫颈癌组织中的表达均高于癌旁正常组织(P<0.01),且AQP5的过表达与宫颈癌的淋巴结转移(P<0.01)及肿瘤分期(P<0.05)有关。ROC曲线结果显示AQP5在宫颈癌组织中的检测敏感性、特异性及AUC值可分别达到72.2%、75.0%和0.715。结论:AQP5在宫颈癌的发生发展中起着重要作用,有机会成为新的宫颈癌的诊断标志物及治疗靶点。
Objective: To investigate the expression of aquaporin 5 (AQP5) in human cervical carcinoma and its clinical significance. Methods: The expressions of AQP5 mRNA and protein were detected by qRT-PCR and Western blotting respectively. The relationship between the expression of AQP5 mRNA and clinicopathological features was analyzed. Using ROC curve to analyze the diagnostic value of AQP5 mRNA in cancer tissue. Results: The expression of AQP5 mRNA and protein in cervical cancer tissues was significantly higher than that in adjacent normal tissues (P <0.01), and the expression of AQP5 was correlated with lymph node metastasis (P <0.01) and tumor stage (P <0.05) )related. The ROC curve showed that the sensitivity, specificity and AUC of AQP5 in cervical cancer tissues were 72.2%, 75.0% and 0.715, respectively. Conclusion: AQP5 plays an important role in the occurrence and development of cervical cancer and has the potential to become a new diagnostic marker and therapeutic target for cervical cancer.
