目的:通过PTU复制大鼠甲状腺肿模型,观察富碘中药复方瘿宁合剂对甲状腺肿大鼠的恢复情况影响。方法:选取4周龄SD大鼠62只,随机选取10只作为正常组,其余52只予以丙硫氧嘧啶(PTU)1 mg/100 g灌胃14 d复制甲状腺肿大鼠模型,随机选取2只大鼠进行模型验证。造模成功后,将50只大鼠按随机对照表随机分为5组,分别为模型对照组、瘿宁合剂1号组、瘿宁合剂2号组、富碘单味药组、碘剂组。瘿宁合剂1号组,予以瘿宁合剂1号浓缩煎剂3 m L灌胃;瘿宁合剂2号组,予以瘿宁合剂2号浓缩煎剂3 m L灌胃;富碘单味药组,予以海藻、海带、昆布浓缩煎剂3 m L灌胃;碘剂组予以碘酸钾溶液3 m L灌胃,以上各组碘含量均为1990μg/m L;模型组予以生理盐水灌胃。记录实验过程中大鼠的体重变化情况,干预28 d后处死大鼠,称取大鼠甲状腺组织重量,进行HE染色观察组织形态。结果:PTU灌胃14 d后,肉眼观察大鼠甲状腺可见其充血肿大,镜下观察可见甲状腺肿形成,甲状腺肿大鼠模型复制成功。与正常组比较,对照组大鼠体重在整个实验过程中呈缓慢增长的趋势,P<0.05;与正常组比较瘿宁合剂1号组、瘿宁合剂2号组、富碘单味药组以及碘剂组体重均呈缓慢增长至增长速度正常的趋势,均P<0.05,各组间比较无统计学差异,P>0.05;镜下观察甲状腺滤泡细胞,富碘中药复方组恢复程度明显好于其他组。结论:PTU能够导致大鼠甲状腺肿大,瘿宁合剂能够使PTU致大鼠甲状腺肿甲状腺滤泡形态得到改善,治疗安全有效。
OBJECTIVE: To investigate the effect of Fuyin Chinese medicine compound Suining Mixture on the recovery of goiter rats by PTU replication rat goiter model. After 28 days of intervention, the rats were killed, the weight of thyroid tissue was weighed, and the morphology of the tissues was observed by HE staining. Results: After 14 days of intragastric administration of PTU, the hyperemia and enlargement were observed in the thyroid gland of rats. The appearance of goiter was observed under the microscope. The rat model of goiter was successfully replicated. Compared with the normal group, the body weight of the control group showed a trend of slow increase throughout the experiment, P <0.05; compared with the normal group, Suining group 1, Suining group 2, iodine single herb group and Iodine group showed a slow increase in body weight to a normal growth rate, both P <0.05, no significant difference between the groups, P> 0.05; microscopic observation of thyroid follicular cells, iodine-rich traditional Chinese medicine compound group was significantly better recovery In other groups. CONCLUSION: PTU can cause goiter in rats. Suining mixture can improve thyroid follicular morphology in rats with PTU, and the treatment is safe and effective.
