目的观察动脉粥样硬化(AS)兔模型中热休克蛋白60(HSP60)的表达及其与斑块稳定性之间的关系和普罗布可对HSP60表达的影响。方法20只新西兰大白兔随机分正常对照组(n=6)、高脂饮食组(n=8)、普罗布可组(n=6)。对照组予以普食喂养,高脂饮食组和普罗布可组予高脂喂养。4周后高脂饮食组和普罗布可组行髂动脉内膜球囊损伤术。术后普罗布可组每只兔子加用普罗布可1000mg/d口服,10周末处死动物。采用HE染色观察动脉病理形态学变化,免疫组化染色检测动脉中HSP60的表达,酶联免疫吸附法测血清中的C反应蛋白(CRP)、白细胞介素(IL)-6、氧化低密度脂蛋白(ox-LDL)、HSP60的水平。结果高脂饮食组动脉可见典型的动脉粥样硬化斑块,普罗布可组无典型斑块形成,内膜增厚较为明显;与高脂饮食组相比较,普罗布可组血清CRP、IL-6、ox-LDL、HSP60水平降低;普罗布可组内膜和中膜HSP60的表达较高脂饮食组明显减少。血清HSP60水平与CRP、IL-6、ox-LDL水平正相关。结论HSP60在兔AS模型中的表达水平明显增加,与斑块的不稳定性有关,普罗布可可抑制兔AS模型血清和斑块中HSP60的表达。
Objective To observe the expression of heat shock protein 60 (HSP60) and its relationship with plaque stability in atherosclerotic rabbits and the effect of probucol on the expression of HSP60. Methods Twenty New Zealand white rabbits were randomly divided into normal control group (n = 6), high fat diet group (n = 8) and probucol group (n = 6). The control group was fed with normal diet, high-fat diet group and probucol group were given high-fat diet. Four weeks later, the high-fat diet group and probucol group received iliac artery balloon injury. After treatment, each group of rabbits was treated with probucol at 1000mg / d orally, and animals were sacrificed at the end of 10 weeks. The pathological changes of arteries were observed by HE staining. The expression of HSP60 in the arteries was detected by immunohistochemistry. The levels of C reactive protein (CRP), interleukin (IL) -6 and oxidized low density lipids Protein (ox-LDL), HSP60 levels. Results The typical atherosclerotic plaque was observed in the arteries of the high-fat diet group and no typical plaque was formed in the probucol group, and the intimal thickening was more obvious. Compared with the high-fat diet group, serum levels of CRP, IL- 6, ox-LDL and HSP60 were decreased. Compared with high fat diet group, the expression of HSP60 in probucol group was significantly decreased. Serum HSP60 levels were positively correlated with CRP, IL-6 and ox-LDL levels. Conclusions The expression of HSP60 in rabbit AS model is significantly increased, which is related to the instability of plaque. Probucol inhibited the expression of HSP60 in serum and plaque of rabbit AS model.
