目的研究重组结核分枝杆菌11000蛋白对结核分枝杆菌感染的鉴别作用。方法以不同分枝杆菌分别致敏豚鼠,致敏成功后给所有动物皮内注射重组结核分枝杆菌11000蛋白和结核菌素纯蛋白衍生物(PPD)或胞内分枝杆菌纯蛋白衍生物(PPD-B),用双盲法记录注射后24h和(或)48h注射局部皮肤反应的纵、横径,计算其均值。结果PPD或PPD-B对所有分枝杆菌致敏豚鼠的皮肤反应均为阳性,局部硬结直径均大于10mm;重组结核分枝杆菌11000蛋白对结核分枝杆菌活菌、牛分枝杆菌、非洲分枝杆菌和堪萨斯分枝杆菌致敏豚鼠的皮肤试验为阳性,局部硬结直径依次为(14.7±2.0)、(9.3±3.8)、(18.7±2.4)和(14.8±4.2)mm,而对灭活结核分枝杆菌、卡介苗和其他非结核分枝杆菌致敏豚鼠的皮肤试验均为阴性。结论重组结核分枝杆菌11000蛋白可有效鉴别结核分枝杆菌活菌感染和卡介苗接种、结核分枝杆菌死菌致敏以及其他非结核分枝杆菌的感染。
Objective To study the differential effect of recombinant Mycobacterium tuberculosis 11000 protein on Mycobacterium tuberculosis infection. Methods The guinea pigs were sensitized by different mycobacteria, and all the animals were intradermally injected with recombinant Mycobacterium tuberculosis 11000 protein and tuberculin pure protein derivative (PPD) or mycobacterium intracellulare pure protein derivative PPD-B). The vertical and horizontal diameters of local skin reactions were recorded by double-blind method 24h and / or 48h after injection. Results PPD or PPD-B showed positive skin reaction to all mycobacterium sensitized guinea pigs, and the diameter of local induration was more than 10mm. The recombinant Mycobacterium tuberculosis 11000 protein had no significant effect on Mycobacterium tuberculosis, Mycobacterium bovis, Mycobacterium and Kansas mcmales sensitized guinea pig skin test was positive, the local diameter of the induration followed by (14.7 ± 2.0), (9.3 ± 3.8), (18.7 ± 2.4) and (14.8 ± 4.2) mm, while the inactivation Mycobacterium tuberculosis, BCG and other non-tuberculosis sensitized guinea pig skin test were negative. Conclusion Recombinant Mycobacterium tuberculosis 11000 protein can effectively identify the Mycobacterium tuberculosis infection and BCG vaccination, Mycobacterium tuberculosis sensitization and other non-Mycobacterium tuberculosis infection.
