目的:探讨丹参滴注液中丹参素钠在Beagle犬体内的药代动力学。方法:健康Beagle犬禁食12 h后,静脉滴注丹参滴注液10 mL.kg-1,血浆样品用乙酸乙酯提取,采用HPLC测定不同时间血药浓度。结果:丹参素钠在0.225~18.000mg.L-1线性关系良好,最低检测限为0.113 mg.L-1,Beagle犬静脉滴注丹参滴注液后的药代动力学参数:tmax为30 min,Cmax为(9.574 2±2.371 5)mg.L-1,t1/2为(19.23±2.97)min,CL为(0.012 7±0.003 0)L.min-1.kg-1,AUC0-tn为(474.954±95.483)mg.min.L-1,AUC0-∞为(482.494±95.353)mg.min.L-1。结论:所建立血药浓度检测方法简便、稳定、可靠,丹参素钠在Beagle犬体内呈单室模型。
Objective: To investigate the pharmacokinetics of danshensu sodium in Beagle dogs in Salvia miltiorrhiza drip. Methods: The healthy Beagle dogs were fasted for 12 hours and then received intravenous drip of Danshen injection (10 mL.kg-1). The plasma samples were extracted with ethyl acetate and the plasma concentrations were determined by HPLC. Results: The linear range of Danshensu sodium was 0.225 ~ 18.000mg.L-1, the lowest limit of detection was 0.113 mg.L-1. The pharmacokinetic parameters of Danshen injection were as follows: tmax was 30 min , Cmax was (9.574 2 ± 2.371 5) mg.L-1, t1 / 2 was (19.23 ± 2.97) min, CL was (0.012 7 ± 0.0030) L.min-1.kg-1 and AUC0-tn was (474.954 ± 95.483) mg.min.L-1 and AUC0-∞ (482.494 ± 95.353) mg.min.L-1. Conclusion: The established method for the determination of plasma concentration is simple, stable and reliable. Danshensu sodium was single-compartment model in Beagle dogs.
