鼠疫菌Evparis株的湿菌体和等量湿菌体制备的菌粉,用2.5%NaCl浸洗可溶性蛋白质,硫酸铵分段沉淀法制备粗制鼠毒素,收获量及其SDS-PAGE电泳图形基本相同。用LB固体培养基增殖鼠疫苗弱毒株,每株培养面积300cm2,收获湿菌体EVparis株0.6g,MⅡ40株、Otten株、A1122株1.2g,175株1.4g。不同菌株每克湿菌体收获粗制鼠毒素的量分别为:MⅡ40株9.34mg,175株14.96mg,EVparis株1575mg,A1122株21.72mg,Otten株26.06mg。粗制鼠毒素SDSPAGE电泳图形显示表征分子量17~94KDa11条蛋白带,不同菌株制备的粗制鼠毒素的蛋白带数量和显色深度均有差异。
The wet bacterial strain of Evparis strain and the bacterial powder prepared by the same amount of wet cells were used to digest soluble protein with 2.5% NaCl and the crude toxin was prepared by ammonium sulfate segregation method. The yield and its SDS-PAGE electrophoresis Graph basically the same. The attenuated strain of mouse vaccine was propagated with LB solid medium. Each cultivating area was 300 cm 2. The wet strain EVparis strain 0.6g, MⅡ40 strain, Otten strain, A1122 strain 1.2g and 175 strain 1.4g were harvested. The amount of crude toxins harvested per gram of wet cells from different strains were 9.34 mg for MII40, 14.96 mg for 175, 1575 mg for EVparis, 21.72 mg for A1122 and 26.06 mg for Otten. The crude toxin SDSPAGE electrophoresis pattern showed that there were 11 protein bands with molecular weights ranging from 17 to 94 kDa. The protein bands and depths of crude toxins prepared by different strains were different.
