目的探讨养阴降压汤(以下称降压汤)对自发性高血压(SHR)大鼠降压作用机制。方法选用雄性SHR大鼠40只,Wistar大鼠10只,随机分为降压汤高剂量组、降压汤中剂量组、西药对照组、空白对照组和Wistar大鼠正常对照组。分别灌胃喂养12周,于第12周末处死大鼠后取出心脏、肾脏和脾脏用实时定量PCR对TNF-αmRNA、IL-10mRNA水平进行检测,同时检测血脂水平、肝功能及肾功能。结果治疗后降压汤高剂量组、中剂量组和西药对照组AST、Cr水平较治疗前显著下降(P<0.01);降压汤高剂量组、中剂量组TC水平较治疗前均有明显下降(P<0.05),高剂量组更为显著(P<0.01)。肾脏和心脏中,降压汤高剂量组、中剂量组和西药对照组TNF-αmRNA水平较对照组明显下降(P<0.05);各组IL-10mRNA水平均无明显变化。结论养阴降压汤可以改善肝肾功能,降低胆固醇,其降压机制可能是抑制了肾脏和心脏中的TNF-αmRNA的表达,使局部TNF-α减少,从而发挥降压和保护肾脏的作用。
Objective To investigate the mechanism of antihypertensive effect of Yangyin Antihypertensive Decoction (SHR) on spontaneously hypertensive rats (SHR). Methods Forty male Wistar rats and ten male Wistar rats were randomly divided into two groups: high-dose-low-dose-Decoction-Tangjiazhuang decoction, medium-dose-Jiangya decoction, western medicine control group, blank control group and normal control group. The rats were sacrificed on the 12th week after the rats were sacrificed. The heart, kidney and spleen were removed. The levels of TNF-αmRNA and IL-10 mRNA were detected by real-time quantitative PCR. The serum lipid, liver function and renal function were also detected. Results After treatment, the levels of AST and Cr in high-dose, middle-dose and western medicine control groups were significantly lower than those before treatment (P <0.01). The TC levels in high-dose and low-dose groups were significantly lower than those before treatment (P <0.05), and the high dose group was more significant (P <0.01). In the kidney and heart, the levels of TNF-α mRNA in high-dose, middle-dose and western medicine groups were significantly lower than those in the control group (P <0.05). There was no significant change in the levels of IL-10 mRNA in each group. Conclusion Yangyinjiangya decoction can improve liver and kidney function and lower cholesterol, and its antihypertensive mechanism may inhibit the expression of TNF-αmRNA in the kidney and heart, decrease the local TNF-α, and thus play a role of antihypertensive and protective kidneys .
