采用分子生物学技术 ,从基因水平上诊断此次术后爆发感染的致病菌。根据分支杆菌的 16srDNA序列 ,设计合成一对引物 ,采用聚合酶链反应技术 ,并以结核分支杆菌、龟分支杆菌龟亚种和偶然分支杆菌做对照 ,对5 3株龟分支杆菌脓肿亚种临床分离株进行PCR扩增。在此基础上 ,采用 16srDNAPCR扩增体系检测 2 5 9份临床标本。结果 5 3株龟分支杆菌脓肿亚种临床分离株均被扩增出一条特异的 5 84bpDNA带。 2 5 9份临床标本 ,PCR扩增阳性率为 62 9%。在基因水平上确认了此次引起院内术后爆发感染的致病菌为龟分支杆菌脓肿亚种
The use of molecular biology techniques, diagnosed at the gene level after the outbreak of infection of pathogens. According to the 16srDNA sequence of Mycobacterium, a pair of primers was designed and synthesized, and polymerase chain reaction (PCR) was used to detect 53 Mycobacterium thaliana abscess subspecies Mycobacterium. Isolates were PCR amplified. On this basis, 259 clinical samples were detected by 16srDNAPCR amplification system. Results A total of 5 84bp DNA bands were amplified from 53 clinical isolates of Mycobacterium turtle abscess. The clinical positive rate of 259 clinical samples was 62.9%.
