目的研究瘦素(leptin)对Hedgehog(Hh)信号通路的调节作用。方法使用3T3-L1细胞诱导脂肪细胞分化,实时定量PCR和Western blot法分别检测脂肪细胞分化过程中Hh信号通路关键转录因子胶质瘤相关癌基因同源物(Gli)mRNA和蛋白的表达,以及leptin对其表达水平的影响。结果在脂肪细胞分化过程中,诱导后的0、4、7、10 d,Gli1、Gli2、Gli3的mRNA和蛋白的表达逐渐受到抑制,以Gli1最为显著;而应用100 ng/m L leptin则可以促进Gli1、Gli2、Gli3的mRNA和蛋白表达。结论在脂肪细胞分化成熟过程中,leptin和Hh具有交互作用,leptin可上调Hh通路信号分子Gli的表达,leptin可能通过激活Hh信号通路发挥抑制脂肪生成作用。
Objective To study the regulatory effect of leptin on Hedgehog signaling pathway. Methods 3T3-L1 cells were used to induce adipocyte differentiation. Real-time quantitative PCR and Western blot were used to detect the expression of Gli mRNA and protein, which are the key transcription factors of Hh signaling pathway, during differentiation of adipocytes.
