目的观察益气养阴祛瘀方和性激素对非肥胖糖尿病(NOD)自发性干燥综合征(SS)样小鼠的影响。方法将60只NOD自发性干燥综合征(SS)样小鼠随机分为模型组、羟氯喹组、益气养阴祛瘀方组、脱氢表雄酮(DHEA)组、雌二醇(E2)组、脱氢表雄酮加雌二醇(DHEA+E2)组,每组10只。ICR小鼠10只为正常组。正常组和模型组给予生理盐水,羟氯喹组给予羟氯喹溶液75 mg/kg,中药组予益气养阴祛瘀方药液27.4 g/kg,DHEA组予DHEA混悬液37.5 mg/kg,E2组给予E2溶液0.375 mg/kg;DHEA+E2组给予DHEA 37.5 mg/kg,E20.375 mg/kg。观察各组小鼠饮水量、唾液分泌量的变化;测定其胸腺、脾脏和颌下腺质量并计算小鼠胸腺指数、脾脏指数和颌下腺指数。酶联免疫法检测血清中细胞因子TNF-α、IL-6水平。结果与模型组比较,各给药组NOD小鼠的饮水量和唾液分泌量均改善(P<0.05)。中药组和DHEA组NOD小鼠的胸腺、脾脏和颌下腺指数均改善(P<0.05)。中药组和DHEA组血清TNF-α、IL-6的水平(P<0.05)显著降低。结论益气养阴祛瘀方及DHEA在整体疗效和免疫功能等方面对NOD小鼠SS具有有效的治疗作用。
Objective To observe the effects of Yiqi Yangyinquyu Decoction and sex hormones on spontaneously non-obese diabetic (NOD) mice. Methods Sixty NOD-SS mice were randomly divided into model group, hydroxychloroquine group, Yiqi Yangyin Quyu Decoction group, DHEA group, estradiol (E2) group ) Group, dehydroepiandrosterone plus estradiol (DHEA + E2) group, 10 rats in each group. 10 ICR mice were normal group. Normal group and model group were given saline, hydroxychloroquine group was given hydroxychloroquine solution 75 mg / kg, Chinese medicine group was given Yiqi Yangyin Quyu Fang liquid 27.4 g / kg, DHEA group was given DHEA suspension 37.5 mg / kg, E2 group was given E2 solution 0.375 mg / kg; DHEA + E2 group was given DHEA 37.5 mg / kg, E20.375 mg / kg. The changes of water and salivary secretion of mice in each group were observed. The thymus, spleen and submandibular gland were measured and the thymus index, spleen index and submandibular gland index were calculated. Serum levels of cytokines TNF-α and IL-6 were detected by enzyme-linked immunosorbent assay. Results Compared with the model group, the water intake and salivary secretion of NOD mice in each administration group were improved (P <0.05). The thymus, spleen and submandibular gland index of NOD mice in Chinese medicine group and DHEA group were all improved (P <0.05). The levels of serum TNF-α and IL-6 in Chinese medicine group and DHEA group were significantly decreased (P <0.05). Conclusions Yiqi Yangyinquyu Decoction and DHEA have an effective therapeutic effect on NOD mouse SS in terms of overall efficacy and immune function.
