目的十二烷基磺酸钠对家蝇幼虫分泌型抗菌肽进行分子鉴定。方法对纯化所得的家蝇幼虫分泌型抗菌肽使用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)及肽指纹图谱方法进行分子鉴定。结果 SDS-PAGE结果表明,纯化得到的2个抗菌组分,主要成分的分子量在14.157 kDa左右,另一组分的分子量为7.579 kDa,但蛋白含量明显低于上一组分;分别将这2个抗菌肽分子命名为家蝇幼虫分泌型抗菌肽Ⅰ和抗菌肽Ⅱ,抗菌肽Ⅰ经肽指纹图谱分析,精确分子量为14.264 kDa,其与溶菌酶序列有一定同源性,序列覆盖率为20%,但两者生物学特性不同,推测为不同蛋白质。结论从家蝇幼虫分泌物中分离得到的抗菌肽与溶菌酶有一定同源性,但生物学特性有差异。
Objective Sodium dodecyl sulfate molecular identification of secreted antimicrobial peptides of housefly larvae. Methods The secreted antimicrobial peptides of the purified Musca domestica larvae were identified by SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and peptide fingerprinting. Results The results of SDS-PAGE showed that the molecular weight of the two antibacterial components was about 14.157 kDa and the other component was 7.579 kDa, but the protein content was significantly lower than that of the previous one. Antimicrobial peptide molecules named housefly larvae secreted antimicrobial peptide Ⅰ and antimicrobial peptide Ⅱ, antibacterial peptide Ⅰ peptide fingerprinting analysis, the exact molecular weight of 14.264 kDa, which has some homology with the lysozyme sequence, the sequence coverage rate of 20 %, But the biological characteristics of the two different, presumed to be different proteins. Conclusion The antibacterial peptides isolated from housefly larvae secretions have some homology with lysozyme, but the biological characteristics are different.
