目的探讨蜘蛛香成分11-ethoxyviburtinal对肠易激综合征(IBS)大鼠肠嗜铬细胞(EC)及肥大细胞(MC)表达的影响。方法 SD雄性大鼠随机分为空白对照组、模型组、匹维溴铵(25 mg/kg)组和蜘蛛香成分11-ethoxyviburtinal大、中、小剂量(1.2、0.6、0.3 mg/kg)组。采用孤养复合慢性不可预见性应激法建立IBS大鼠模型,免疫组化法检测大鼠结肠5-羟色胺(5-HT)免疫反应阳性细胞EC数量的变化,甲苯胺蓝染色法检测大鼠结肠MC数量的变化。结果孤养复合慢性不可预见性应激可造成大鼠结肠5-HT免疫反应阳性细胞EC数量显著增加(P<0.01),结肠MC数量显著增多(P<0.05)。蜘蛛香成分11-ethoxyviburtinal大剂量可显著减少结肠5-HT免疫反应阳性细胞EC和MC数量(P<0.05)。结论蜘蛛香成分11-ethoxyviburtinal可抑制IBS大鼠结肠EC和MC的表达。
Objective To investigate the effects of spider incense 11-ethoxyviburtinal on the expression of enterochromaffin (EC) and mast cells (MC) in rats with irritable bowel syndrome (IBS). Methods SD male rats were randomly divided into blank control group, model group, piroxicam (25 mg / kg) group and spider incense components 11-ethoxyviburtinal large, medium and small doses (1.2,0.6,0.3 mg / kg) . IBS rats were established by loneliness combined with chronic unpredictable stress. The changes of EC number of 5-HT (5-HT) immunoreactive cells in the colon of rats were detected by immunohistochemical staining. Toluidine blue staining Changes in the number of colon MC. Results Lonicera complex chronic unpredictable stress could cause the number of colon 5-HT immunoreactive positive cells in colon increased significantly (P <0.01), and the number of colonic MC significantly increased (P <0.05). The high dose of 11-ethoxyviburtinal could significantly decrease the number of EC and MC in the colon 5-HT immunoreactive cells (P <0.05). Conclusion Spider incense component 11-ethoxyviburtinal can inhibit the colon EC and MC expression in IBS rats.
