用反相高效液相法,对血浆非那西汀药物浓度的测定方法及其药代动力学进行了研究。实验采用Waters公司HPLC系统,国产ZORRAXBPCN柱,以磷酸盐缓冲液/乙氰为流动相,内标法定量。本方法标准血浆线性关系良好(r=09999),线性范围070~8928μmol/L,最低检测量为125ng,平均方法回收率为9992%,浓度为8928μmol/L、1116μmol/L的提取回收率分别为948%、831%和824%;精密度考察批内变异系数(CV%)分别为049、365和490,批间变异系数(CV%)分别为393、364和593。
RP-HPLC method was used to determine the concentration of phenacetin in plasma and its pharmacokinetics. Experiments using Waters HPLC system, made ZORRAX BP column, with phosphate buffer / acetonitrile as the mobile phase, internal standard method. The standard linearity of this method was good (r = 0.9999) with a linear range of 070-8928μmol / L, with the lowest detection level of 125ng. The average recovery was 9992% with a concentration of 8928μmol / L and 1116μmol / L were 948%, 831% and 824%, respectively. The coefficient of variation (CV%) within the precision was 0.49, 3.65 and 490, the inter-assay coefficient of variation (CV%) were 393,364 and 593, respectively.
