目的:制备羟基喜树碱磷脂复合物,进行溶解度考察和表征,推测其形成机理并为下一步的制剂制备提供参考。方法:以四氢呋喃为复合溶剂制备羟基喜树碱磷脂复合物,考察磷脂复合物在正辛醇与氯仿2种溶剂中的溶解性能,并采用紫外(UV)、红外(IR)与差示扫描量热法(DSC)对复合物进行表征。结果:羟基喜树碱磷脂复合物在氯仿中的溶解度显著提高但不稳定。紫外、红外图谱与DSC结果验证了复合物的形成。结论:羟基喜树碱可能与磷脂的极性端复合形成复合物,药物的脂溶性与溶出速率得以提高。
OBJECTIVE: To prepare hydroxycamptothecin phospholipid complex for investigation and characterization of its solubility, speculate its formation mechanism and provide a reference for the preparation of the next step. METHODS: Hydroxycamptothecin phospholipid complex was prepared with tetrahydrofuran as solvent. The solubility of phospholipid complex in two solvents of n-octanol and chloroform was investigated. The UV-vis, infrared (IR) and differential scanning The complex was characterized by thermal analysis (DSC). Results: The solubility of hydroxycamptothecin phospholipid complex in chloroform was significantly increased but not stable. UV, IR and DSC results confirmed the formation of complexes. CONCLUSION: Hydroxycamptothecin may complex with the polar end of phospholipid to form a complex, and the liposolubility and dissolution rate of the drug may be increased.
