目的:通过观察实验性精索静脉曲张大鼠睾丸生精细胞凋亡指数(AI)、睾丸组织Caspase-3,Hsp60表达变化,探讨实验性精索静脉曲张大鼠睾丸生精细胞凋亡状况及通精灵对精索静脉曲张大鼠睾丸生精功能的保护作用。方法:选择成年清洁级雄性W istar大鼠50只,按Saypol法制作左精索静脉曲张大鼠模型,随机分为假手术组10只、模型组10只、通精灵低剂量组10只、通精灵中剂量组10只、通精灵高剂量组10只。造模1个月后,通精灵各组予以不同浓度的通精灵水煎剂,通精灵各组均每日1次灌胃。假手术组和模型组予以等量的生理盐水灌胃,每日1次,连续2个月。各组实验大鼠普食喂养2个月后,全部处死,取出睾丸,用原位末端脱氧核苷酸转移酶标记法(TUNEL)检测睾丸生精细胞凋亡,免疫组化法检测睾丸组织Caspase-3、Hsp60表达。结果:①模型组大鼠左睾丸凋亡指数明显高于假手术组及通精灵各组(P<0.01),通精灵各组左睾丸凋亡指数与模型组比较,差异有显著性(P<0.05)。②模型组大鼠睾丸组织Caspase-3、Hsp60蛋白表达上升;与假手术组比较,差异有显著性(P<0.01,P<0.05),通精灵中、高剂量组Caspase-3、Hsp60蛋白表达显著下降,与模型组比较,差异有显著性(P<0.05)。结论:通精灵能降低实验性精索静脉曲张大鼠睾丸生精细胞凋亡,减轻实验性精索静脉曲张大鼠睾丸组织的病理损害;通精灵可下调Caspase-3、Hsp60表达,这可能是通精灵抑制生精细胞凋亡,改善精索静脉曲张不育症患者生精功能机制之一。
Objective: To investigate the apoptosis of spermatogenic cells in experimental varicocele rats by observing the apoptosis index (AI) of testicular spermatogenic cells and the expression changes of Caspase-3 and Hsp60 in experimental testicular varicocele rats. The Protective Effect of Tongjie Elders on Testicular Spermatogenesis in Varicocele Rats . METHODS: Fifty male adult Wistar rats were selected and the left varicocele rat model was made according to the Saypol method. The rats were randomly divided into 10 sham-operated groups, 10 model groups, and 10 low-dose groups. There were 10 middle-dose groups and 10 high-dose groups. One month after the modeling, each group of the elves was given a different concentration of the decoy water decoction. Each group of elves was fed one time a day. The sham group and the model group were given intragastric administration of equal volume of saline once a day for 2 months. Each group of experimental rats was fed for 2 months and then all were killed. The testes were removed and the apoptosis of spermatogenic cells was detected by in situ terminal deoxynucleotidyl transferase labeling (TUNEL). The testicular tissue was detected by immunohistochemistry. -3, Hsp60 expression. Results: 1 The apoptotic index of the left testis in the model group was significantly higher than that in the sham-operated group and the Tongjingling group (P<0.01). Conclusion: Tong Jingjing can reduce the apoptosis of testicular spermatogenic cells in experimental varicocele rats and reduce the pathological damage of testicular tissue in experimental varicocele rats; Tongjieling can down-regulate the expression of Caspase-3 and Hsp60, which may be Tong Jingling inhibits apoptosis of spermatogenic cells and improves one of the mechanisms of spermatogenesis in patients with varicocele infertility.
