目的采用超高效液相色谱法(UPLC)快速测定甲钴胺片的含量。方法采用Agilent Zorbax Plus C_(18)色谱柱(50 mm×3.0 mm,1.8μm),以乙腈-水为流动相,梯度洗脱,流速0.8 m L·min~(-1),检测波长267 nm,柱温30℃,采集频率80 Hz,进样体积3μL。结果甲钴胺片13.95~25.91μg·m L~(-1)与峰面积呈良好的线性关系(r=0.9999),平均回收率为100.0%,RSD=0.8%(n=9)。结论所用方法简便、准确、专属性强,可用于甲钴胺片的含量测定。
Objective To rapidly determine the content of mecobalamin tablets by ultra performance liquid chromatography (UPLC). Methods The Agilent Zorbax Plus C_ (18) column (50 mm × 3.0 mm, 1.8 μm) was used as the mobile phase. The mobile phase consisted of acetonitrile and water with a flow rate of 0.8 m L · min -1. The detection wavelength was 267 nm , Column temperature 30 ℃, acquisition frequency 80 Hz, injection volume 3μL. Results The average linear range was 13.95 ~ 25.91 μg · m L ~ (-1) and the peak area was (r = 0.9999). The average recovery was 100.0% and RSD was 0.8% (n = 9). Conclusion The method is simple, accurate and specific and can be used for the determination of mecobalamin tablets.
