目的探讨麦芽酚铝[aluminum maltolate,Al(mal)_3]染毒对大鼠肾上腺嗜铬细胞瘤细胞(rat pheochromocytoma cells,PC12)钙稳态及凋亡的影响。方法将PC12细胞随机分为0(对照组)、50、100、200、400μmol/L Al(mal)_3染毒组,分别培养12、24、48 h,采用CCK-8试剂盒检测PC12细胞活性,以流式细胞术检测PC12细胞内游离钙离子荧光强度[Ca(~2+)]i及细胞凋亡率。结果随着Al(mal)_3染毒时间和染毒剂量的增加,PC12细胞活性逐渐下降,细胞[Ca(~2+)]i与细胞总凋亡率逐渐升高。除50μmol/L Al(mal)_3染毒12、24 h及100μmol/L Al(mal)_3染毒12 h外,其他各剂量组染毒PC12细胞12、24、48 h后细胞活性均低于对照组,细胞[Ca(~2+)]i均高于对照组,差异有统计学意义(P<0.05)。400μmol/L Al(mal)_3染毒PC12细胞12 h,200、400μmol/L Al(mal)_3染毒24 h,100、200、400μmol/L Al(mal)_3染毒48 h后,PC12细胞总凋亡率均高于对照组,差异有统计学意义(P<0.05)。结论钙超载可能是Al(mal)_3引起凋亡发生的主要机制。
Objective To investigate the effects of aluminum maltolate (Al (mal) 3] on calcium homeostasis and apoptosis in rat adrenal pheochromocytoma cells (PC12). Methods PC12 cells were randomly divided into 0 (control group), 50,100,200,400μmol / L Al (mal) _3 group and cultured for 12,24,48 h respectively. Except for 12 and 24 h of exposure to 50 μmol / L Al (mal) 3 and 12 h of exposure to 100 μmol / L Al (mal) 3, the cell viability of PC12 cells was lower than that of the control Control group, cell [Ca (~ 2 +)] i were higher than the control group, the difference was statistically significant (P <0.05). PC12 cells were exposed to 400 μmol / L Al (mal) 3 for 12 h, exposed to 200, 400 μmol / L Al (mal) 3 for 24 h, exposed to 100, 200, 400 μmol / L Al The total apoptosis rate was higher than the control group, the difference was statistically significant (P <0.05). Conclusion Calcium overload may be the main mechanism of Al (mal) _3 apoptosis.
